In the end, Donatello concludes that no, it was entirely impossible for them to have turned to humans. They were originally turtles, how would they’ve been able to become one? If they were human, who’d be the mother? Would their personalities alter if that were to occur? Would everything alter?
But Donnie should’ve known that in their world, anything was entirely possible.

Casey had never felt less prepared for what was ultimately going to be a mundane evening. He was meeting Donnie's family. The Hamato Family. Palms sweaty mom's spaghetti.
...He buried his face into his shoulder again and Donnie let him. A hand coming up to smooth through his hair. “Are you really that nervous to meet them?”
“Casey Jones ain’t afraid of nobody.”
“Uh huh.”
“I’m a little nervous. I’m just overthinking it is all.”

Donnie's life is composed of two important factors: his academics, and his favorite podcast. His drive to remain at the top of his school and to get accepted into the most prestigious of colleges swallows up every aspect of his life until it's the only thing remaining, aside from his love of the one podcast that's been there for him when everything else has morphed and changed.
A deal to take more chances leads Donnie on a journey that he never expected to have. It's the type you always read about in books, but never the type you expect to happen to you. And it all begins because Donnie took a chance and simply said "Yes."

Summary
There was something weird about the family that just recently moved in next door. First of all, the new neighbor had four kids, quadruplet, and while Yoshi had met all the adorable little boys, he’d almost never seen any adults watching over them. Second, the boys were wise, way too mature for their age, it made Yoshi wondered what exactly they had gone through. Third, they all acted like they had known him for a very long time.
(Or in which, one day, the turtle brothers woke up as human boys in a reality different from their own.)
