During episode 5x19. Lena gets jealous when Kara worries about William and realises that she has feelings for Kara. She spends the evening at Kara's apartment talking, drinking and by the end of the night telling Kara how she really feels about her. Lena ends up staying the night.
Basically the supercorp endgame we were waiting for but never got at the end of that episode.
