Hearing that Cas had ignored Sam’s call was the first major sign that something was cosmically wrong. There were minor signs of course, and all piled together they were impossible to ignore.
Sam was different, and not in any predictable sense.

John Winchester kept some things close to his heart. He was a hardened man with only one goal and focus in mind, to avenge Mary's death. But in order to do that, John was willing to sacrifice anything and everything, even giving up the other six-month old son that he had for adoption.
Dean Winchester had lots of memories. Memories of his mom, memories of his brother, and dreams of what his life could have been if they were just alive. But no, it was just him, dad, and Baby. And a bunch of dirty motels in between.
Ben learned to never need or ask for anything, but there was just one thing he always wanted: to be loved. To be cared for and to be loved by an actual family. But he was just a slave, trapped into the realm of monsters and hunters because Ben was a monster, a psychic, and after getting a vision of the yellow-eyed demon, was now placed into the hands of the Winchesters.

Everything was normal again... except for Sam’s memory, that was. Which happened to be... well, Dean thought it sucked. Sam was just growing tired.
In the second part of this series, we get to see more of the boys’ day-to-day life along with Sam’s new condition — still heavy with angst and hurt/comfort.
